The Amazing Spider Man tetap berceritakan seorang nerd bernama Peter Parker yang mendapatkan kekuatan super dari gigitan seekor laba - laba bedanya ternyata laba - laba ini adalah hasil mutasi gen yang dilakukan oleh perusahaan oscorp, perusahaan yang ternyata pernah menaungi ayah peter parker. Satu lagi yang beda disini, tidak ada kisah tentang Mary Jane melainkan menceritakan pertemuan si Peter dengan Gwen Stacy yang pernah di ceritakan di Spiderman versi Sam Raimi.
